MoBiTec Expands Its Selection of High Quality Gene Expression Systems for Bacillus megaterium

Based on its already successful protein expression systems for Bacillus megaterium, MoBiTec GmbH introduces a new family of protein expression vectors, the hp-Expression Systems, for high yield expression in the well-established organism Bacillus megaterium. This new system complements the existing non-E. coli expression systems for B. megaterium, Bacillus subtilis, Lactococcus lactis and yeast. Further non-E. coli expression systems are in preparation.

Goettingen, Germany, June 09, 2011 --(PR.com)-- MoBiTec GmbH, leading provider of non-Escherichia coli protein expression systems, expands its series of expression vector systems suitable for protein expression in Bacillus megaterium.

Bacillus megaterium is a well established and preferred host for heterologous gene expression and an excellent alternative to the widely used working horse Escherichia coli. B. megaterium is a widely used host for gene expression and does not produce toxic compounds like lipopolysaccharides which makes the purification of produced proteins less tedious and time consuming. The protein yield obtained with the optimized high performance hp vector systems is up to 10 times enhanced compared to protein expression with the respective basic plasmid.

hp vectors are available for intracellular expression, without or with affinity tag for convenient purification:

· pC-His1623hp, E. coli/B. megaterium shuttle vector, C-terminal 6xHis-tag
· pN-His1623hp, E. coli/B. megaterium shuttle vector, N-terminal 6xHis-tag
· p3STOP1623hp, E. coli/B. megaterium shuttle vector

as well as for secretion with 2 different signal peptides:

· pSPLipA-hp, E. coli/B. megaterium shuttle vector with signal sequence
· pSPYocH-hp, E. coli/B. megaterium shuttle vector with signal sequence

Bacillus megaterium protoplasts specifically optimized for transformation

The protoplasts supplied by MoBiTec are from B. megaterium strain WH320, developed by Prof. Dr. W. Hillen at the Institute of Microbiology in Erlangen, Germany. These protoplasts are prepared according to an optimized protocol resulting in the highest transformation efficiencies.

For secretion vectors they offer protoplasts of strain MS941. Both strains are mutants of DSM319, whereby WH320 is a chemically mutant, while MS941 has a defined deletion in the gene of the major extracellular protease NprM.
