MoBiTec Launches SpeedBlot (His) – A Strikingly Fast and Easy-To-Use Western Blot Stain for His-Tagged Proteins

Instant detection of poly-histidine tagged proteins on nitrocellulose membrane with SpeedBlot (His) is easy and does not require relevant hands-on time. Even 0.1 pmol detection limits can be achieved with this highly sensitive detection reagent. While common Western Blot protocols require 6 to 21 hours, staining with SpeedBlot (His) is completed after just about 90 minutes.

Göttingen, Germany, June 12, 2014 --( Western Blot is widely used in research to separate and identify proteins. In this technique a mixture of proteins is separated based on molecular weight through gel electrophoresis. The content of the gel is then transferred to a membrane producing a band for each protein. The protein of interest is then detected by means of a specific antibody.

Poly-histidine tags are often used for affinity purification of poly-histidine-tagged recombinant proteins expressed in Escherichia coli, Bacillus megaterium, Bacillus subtilis, and other prokaryotic expression systems. A poly-histidine tag is an amino acid motif in proteins that consists of multiple histidine (His) residues, mostly located at the N- or C-terminus of the protein. In order to facilitate the detection of successfully expressed his-tagged proteins the usage of anti-poly-histidine antibodies is a prevalent technique.

However, the common protocol for the antibody-based detection of the protein of interest is a laborious and time-consuming undertaking.

As time is money and to make life easier MoBiTec introduces SpeedBlot (His), a ready-to-use reagent for the detection of poly-histidine tagged proteins in blot assays:

SpeedBlot (His) is equally suitable for Western Blot assays and Dot or Slot Blot assays. The detection reagent is made of an optimized anti-histidine antibody gold nanoparticle conjugate and is reactive against C- and N-terminal tags. The protein of interest is specifically stained in pink directly on the blot.

As Easy as Ponceau Red but as Specific as ECL Antibody!

• Very fast protocol; results in 30-90 min
• Specific staining of His-tagged proteins
• Sensitive to sub-picomol amounts of protein
• Simple incubation, no buffer exchange, etc.
• Economic, no readout equipment required
• Visually detectable result
• Quantitative and permanent signal
• Works with C-/ N-terminal tags
