With violet light, single cells in an EosFP marked (green) cell population can be switched to red, so that their fate can be followed selectively, for example in embryonic cell differentiation. Alternatively, a particular compartment within an EosFP marked (green) cell can be switched to red.

The Green-to-Red switchable, photochromic Fluorescent Protein „EosFP“ offers advantageous applications in Molecular Cell Biology, and is now available through the German biotech company MoBiTec GmbH.

The Green-to-Red switchable, photochromic EosFP protein, isolated from the stone coral Lobophyllia hemprichii, normally fluoresces green, similar to the well-known Green Fluorescent Protein (GFP, Nobel price 2008). As a particular additional property, violet light (390 nm) can switch the emission maximum of EosFP from green (516 nm) to red (581 nm). Due to this property, the protein is named after the Greek goddess of the dawn, Eos. The violet light induces a photo dissociation of the peptide backbone and is thus irreversible. In its natural form, EosFP is a tetramer; the protein was however genetically modified so that dimeric and monomeric mutant proteins are available. EosFP fusion proteins were shown to keep their biological activity.

With violet light, single cells in an EosFP marked (green) cell population can be switched to red, so that their fate can be followed selectively, for example in embryonic cell differentiation. Alternatively, a particular compartment within an EosFP marked (green) cell can be switched to red allowing then to specifically observe the properties of these now red molecules within the green cell. Red marked proteins offer advantages in cell biology since they can be irradiated with low-energy and thus less harmful light, and because longer wavelengths cause less background due to lower levels of light scattering and autofluorescence.

Developed at the University of Ulm, Germany, MoBiTec GmbH obtained the rights for commercialization of this exciting new tool for life science research.
Two variants of EosFP are available from MoBiTec: The tetrameric wild type protein (EosFP) and a pseudomonomeric variant in which two copies of an engineered EosFP variant are fused to form a tandem dimer (td-EosFP). Both variants express functionally in a wide range of pro- and eukaryotic cells at a temperatures of 37°C or below. For the labeling of cells or tissues, tetrameric EosFP is the construct of choice. For labeling of subcellular compartments using short oligopeptide signals attached to the marker, both EosFP and td-EosFP can be considered. Although some fusion proteins with tetrameric EosFP are possible, the pseudomonomeric variant td-EosFP is the recommended construct for protein labeling. Fusions to the N-terminus of td-EosFP usually work well. Fusions to the C-terminus are also possible, however, some fusion might fail with proteins requiring a strictly monomeric marker, for instance tubulin.
